Peer-Reviewed Literature & Media Analysis
Dr. Patrick E. Sewell's foundational framework on DNA damage response overactivation as an integrative driver of biological aging.
The Guardian Paradox proposes that chronic, largely mutation-independent overactivation of the DNA damage response (DDR) dysregulates the cell's intact guardian pathways—the tumor-suppressor networks and their negative regulators—to induce senescence, deplete stem-cell pools, and drive inflammaging. Unlike cancer, where mutations inactivate these guardians, aging more often reflects functional dysregulation of wild-type pathways (p53, p16INK4a, SIRT1, FOXO3, NRF2, Klotho).
Featured below are the landmark 2026 paper published in the journal Aging and the in-depth investigative feature by Science 2.0 examining this evolutionary double-edged sword.
The Guardian Paradox (Aging Journal Review Paper)
Published August 31, 2026. Organizes 16 candidate aging axes across three evidence tiers with conceptual diagnostic signatures and pre-specified falsification tests.
Science 2.0 Feature Article (Genetics & Molecular Biology)
Published September 21, 2026. Analyzes how nature's early-life anti-cancer surveillance inadvertently drives post-reproductive senescent SASP inflammatory storms.
The Guardian Paradox Definition
“That the very programs guarding against cancer can, when chronically engaged, come to drive aging we term the guardian paradox.” — Patrick E. Sewell, MD (Aging, Aug 31, 2026)
